Following are the steps to calculate stomata per mm2:
i) On a leaf peel that has been properly stained and cut, under the microscope, locate an area that has excessive number of stomata and one which does not have any other hindrances such as finger impressions, etc.
ii) Count the number of stomata here. Locate three more such areas.
iii) Add all the data together and divide by four to determine an average.
iv) Now you can calculate the number of stomata per mm2 by multiplying by 8.
Note that the microscope should be set at 400X.
