Slowly add the alcohol to dissolve the powder. Add 30 mL saturated alcoholic solution of methylene blue to 100.0 mL distilled water and 0.1 mL 10% potassium hydroxide. Filter this solution and dilute it 1:20 with distilled water to prepare the final methylene blue counterstain.
