STEPS TO OBSERVE THE CELLS IN ONION PEEL UNDER A MICROSCOPE:
- Onions fall under the genus cotyledons
- Onions are made up of fleshy layers that are divided by a thin membrane.
- For this experiment, we will use the membrane which can be easily obtained by removing it with the help of tweezers
To prepare the wet mount slide:
- Add a tiny drop of water at the center of the slide
- Put the onion membrane over the drop of water.
- stain the membrane with methylene blue or iodine
- cover the slide with a cover slip with a help of tweezers making sure to avoid any trapping of air bubbles.
- Place the prepared slide under a microscope and observe.
OBSERVATION:
- Cells are linear, individual and rectangular shape.
- The size of each cell ranges from 0.25 to 0.4 meters in length.
- The cell are surrounded by cell wall which helps the cell in retaining their distinct shape.
- A distinct stained nucleus is present that is positioned towards the periphery of cell.
- A large vacuole is present at the centre of the cell
