STEPS TO OBSERVE THE CELLS IN ONION PEEL UNDER A MICROSCOPE:

  • Onions fall under the genus cotyledons
  • Onions are made up of fleshy layers that are divided by a thin membrane.
  • For this experiment, we will use the membrane which can be easily obtained by removing it with the help of tweezers

To prepare the wet mount slide:

  • Add a tiny drop of water at the center of the slide
  • Put the onion membrane over the drop of water.
  • stain the membrane with methylene blue or iodine
  • cover the slide with a cover slip with a help of tweezers making sure to avoid any trapping of air bubbles.
  • Place the prepared slide under a microscope and observe.

OBSERVATION:

  • Cells are linear, individual and rectangular shape.
  • The size of each cell ranges from 0.25 to 0.4 meters in length.
  • The cell are surrounded by cell wall which helps the cell in retaining their distinct shape.
  • A distinct stained nucleus is present that is positioned towards the periphery of cell.
  • A large vacuole is present at the centre of the cell
