When it comes to bateria, there are two basic forms of cell walls: gram-positive and gram-negative. Gram positive cells have a thick peptidoglycan layer. This thick layer retains the crystal violet stain (purple color) when stained. Gram negative organisms have a thin peptidoglycan layer and an overlying outer membrane. These stain pinkish due to the counter-stain used in the Gram staining procedure. It basically comes down to the composition of the cell wall.
