To get 2 X10>6 monocytes from human blood, you will need to draw at least 5 ml of blood. However, if you need to freeze the monocytes and later will thaw it, you will need more monocytes for storage, because freezing and thawing will kill some cells. So, it is always better if you took 12 ml of blood, isolate the PBMC first, then sort the monocytes by using CD14 Magnetic beads. This method will give you at least 5X10>6 monocytes.
