how to observe onion root tip​

  • The root tips was transfered in 1N Hydrochloric acid and incubate at 60°C for 5 minutes. Stained with acetocarmine for 10 minutes. Transferred onto a clean slide and covered with a cover slip. Gently tap the root and observe under the Microscope and Foldscope.

onion root tip, cover slip, slide, filter papers, glycerine, microscope.

➜Take an onion root tip.

➜Place it on slide.

➜Put a drop of water and then add a drop glycerine of water.

➜Cover it with cover slip.

➜Put two or three layers of filter papers on cover slip.

➜Tap the cover slip and gently press the slide to spread the material.

➜Observe under the microscope cope.
