At night, or in the absence of light, photosynthesis in plants stops, and respiration is the dominant process. The plant uses energy from the glucose it produced for growth and other metabolic processes.

The "light-independent" or dark reactions happen in the stroma of the chloroplasts.

These processes can only happen in the chloroplast (a chlorophyll filled plastid in green plants), photosynthesis can only happen in green plants.
