You want to calculate the percentage of free haemoglobin /total Hb right?
Then you have to consider that total Hb in your sample is present in 2 fractions: i. in the packed red blood cells and ii. in the supernatant (free Hb resulting from haemolysis). The packed RBCs represent a given percentage of the total volume of your sample and this percentage is the Ht. Similarly, the supernatant represents a given percentage of the total volume of your sample and this percentage is 100-Ht, right. Since you measure free Hb concentration in the supernatant you have to take into account that the supernatant is only 100-Ht% of the total volume. The formula you have found makes the appropriate correction to appreciate free Hb in the total volume.
