Boil the leaf in water.
Now put it in some methylated spirit and keep the methylated spirit beaker in a boiling water. This will completely denatured the enzymes and kill the cells. Now wash the leaf, and pour a few drops of Iodine Solution on the leaf. The areas with starch will turn blue- black in colour, whereas the areas without starch will remain the same as before (pale white in colour)
