Hypothetically assume a plane in moving form N to S and the earth is moving from S to N.Will their possibly be any change in distance as well as time for the plane to reach the desired location.(Earth is moving at it's actual speed)

Nope. Because speed is a relative term. So when we talk about speed than, unless anything is mentioned, it means speed relative to Earth. So it doesn't matter whether Earth is moving or not. We consider it stationary.
