2 squared is 4. If this is the “first” square number, then you’re ignoring the squares of fractions and irrational numbers, you’re just squaring integers, and you’ve left out the square of 1 for some reason.

But that means we need the square of the eleventh integer, which is to say 11.

11*11=121

Now it still nags at me that you left out 1, the square of 1. But 1 is no longer considered a prime number. What if your definition of squares here rules out all non prime number squares? The first prime is 2, so the first square of a prime is 4.

This means we’d need the tenth prime, rather than the 11th integer. 29 is the tenth prime, and 841 is the square of 29, so 841 is the 10th square of a prime.

But I doubt that’s what you meant, it’s just… hard to tell without more details.
