if a large number of account holders do not wish to keep their money in the bank, how will it affect the banks working?

it only not effect the banks but it also effect our country economy as banks also have to contribute to our GDP

It will effect bank as Bank work on the principle that they take money from those who has sir surplus and gives them on rent who wants money but if no one wants to keep their money in bank then they would not be able to exchange loans and hence they would not get any profit.
