- First of all, a normal sodium chloride solution is also known as a saline solution; it contains 0.85% NaCl in the water while PBS buffer contains 0.9% NaCl in Phosphate buffer having KCl.
- The use of buffers depends on what tissue or where we are using it or in simple words its site of application; usually, we use PBS buffer in medical researches and surgeries to keep organ alive and maintain its physiological pH, whereas sodium chloride solution is used in laboratories with adjusted tonicity.
- We cannot use NaCl solution directly during insulin extraction since it has tonicity different (maybe hypertonic/hypotonic) than the cells in our tissue that may result in cell lysis.
- On the other hand, we can use PBS buffer directly since it is isotonic in nature and does'nt lead to cell lysis.
