The potted plant is kept in a darkness for few time so that all the starch stored in it is used by the plant and no more food in the form of starch is present in it.
Alcohol is used because alcohol absorves all the chlorophyll present in a leaf and by observing chlorophyll the leaf fades in colour or little bit colourless and the all the green colour of the leaf transferred to the alcohol solution and alcohol solution becomes green in colour.
