In experiment to prepare the temporary mount of a leaf peel to show stomata..we use glycerine to moist the leaf so that it guard cell can't be closed..or we use safranin because it is a stain which help us see clearly the cell in the stomata...

This highlights the cells while visualising under a microscope whereas the glycerine is used to keep the stomata wet in order to facilitate staining and viewing in the microscope, this works as a moisturiser and also protects the internal parts of the leaves.
