With repetition means that something that has been used can be used again.

Without repetition means that once something is used, it cannot be used again.

For example, think about those problems where you do something like make three letter "words" from the letters of "HORSE".

With repetition, we include words like HOH, RRR, SEE, and so on, as well as things like SHE, REH, of course.  In total, there are 5³ = 125 different words.

Without repetition, only the words like SHE, REH, and so on, are allowed.  The ones like HOH, RRR, SEE are not permitted.  A letter can only be used once.  The total number of words in this case is 5×4×3 = 60.
