“Despite all her efforts, Susan did not succeed.” The word in spite is always followed with a preposition “of” however, the word despite is never followed with a preposition, i.e. “of.”

This helps in keeping the meaning of the sentence constant without altering, although, the initial word changes to “despite” instead of ”in spite of.” Both words have similar meaning expressed in a different way.
