a. The leaf is dip in boiling water for a minutes to kill the cells. And it destroys enzymes in the protoplasm, prevents any chemical changes in the leaf.

b. Boil the leaf in methylated spirit over a water bath till it become pale-white beacuse of loss of chlorophyll. And it make the colour changes more clear during idoine test.

c. It turns blue-black if starch is present and stains brown if starch is absent.
