Is callus cell highly differentiated or not?

No,callus is a mass of undifferentiated cells having totipotent cells.
It is made to differentiate by application of cytokinins(shoot differentiation) and auxins(root differentiation) along with other hormones and minerals in tissue culture method of plant propagation techniques.
