Plasmotomy is a type of sexual reproduction in which a miltinucleate protozoan cell divides into two or more miltinucleate daughter cells.
In multiple fission the nucleus of the parent cell divides several times by amitosis. So they're different

in multiple fission nucleus divides several times and multinucleate daughter cells are formed through encystment.

in plasmotomy multinucleate daughter cells are formed without the formation of cyst and nucleus do not divide.

thus they both are same in respect of multinucleate daughter cells.
