it all depend on situations
if you have all the qualities of a true friend you will definitely deny when your friend is doing something wrong or he or she is going on a wrong path, or maybe he or she is taking wrong decision bcoz if you will not stop or say no to her or him at that time, something wrong will definitely happen, and you can do nothing at that time instead of regretting
somerimes no matter how true you are some situation puts us into dilemma, whether to say yes or no

Absolutely. Friends understand that their friends have their own lives, their own situations, and their own moods. You each have families, and other friends, and maybe jobs or other responsibilities which means sometimes you must say no. Even if you want to help, sometimes you can't or shouldn't.
Also, if a friend asks you to keep a secret, it's usually good to say you can't promise that, because if keeping the secret means someone gets hurt (as in, your friend wants to commit suicide or knows someone who wants to) then you would feel that you had to respond, secret or not.
Also, if someone asks you to do something illegal or immoral, then (unless someone's life is in danger) that person is either not your friend or has a lot of apologizing to do.
