First remove stomatal peel of a leaf carefully. Then color it by adding saffranine for better observation of leaf peel. Clean leaf peel if extra amount of saffranine is added. Then keep the peel on the glass slide and add some drops of glycerine and cover it with a cover slip.

1. take the lower epidermis of leaf peel.

2. peel of the leaf from its undersurface and put the pea in the watch glass containing water .

3. after add a few drops of safranin stain into the watch glass to stain the peel .

4. after staining put the peel on the clean slide and cut it in a rectangular or square shape .

5. with the help of a filter paper remove the excess of stain in water .

6.put a drop of glycerine on the slide over the peel as mounting chemical gently put the cover sleep to avoid the entry of air bubbles and observe under the microscope .
