Hi my dear friend,

First remove stomatal peel of a leaf carefully. Then color it by adding saffranine for better observation of leaf peel. Clean leaf peel if extra amount of saffranine is added. Then keep the peel on the glass slide and add some drops of glycerine and cover it with a cover slip.

•Clean leaf peel if extra amount of saffranine is added.

•Then keep the peel on the glass slide and add some drops of glycerine and cover it with a cover slip.

Make sure that , the oxygen bubbles are not inside.

•Now the slide can be observed under microscope..
