Mechanism and factors influencing dermal penetration of drug by queras
Answers
Dermal penetration testing is followed to measure the absorption of substance through the skin barrier. It is also known as percutaneous penetration that is conducted to determine how much of a substance (Mostly chemicals) are penetrated through the skin.
The test is also conducted to determine the potentiality of the skin over the substance to absorb it further to the systematic circulation.
As like dermal penetration test dermal toxicity test is conducted to acknowledge the systematic effect of the chemical soon after the dermal penetration.
Administration of drug through skin is called trans-dermal administration.
Mechanism of drug penetration
1. The hair follicles with their associated sebaceous glands,
2. The the sweat ducts;
3. ( Transcellular or Intercellular) across the continuous stratum corneum between these appendages
Factors influencing drug absorption through this route are
- Skin hydration.
- Temperature and pH.
- Diffusion coefficient
- Drug concentration
- Partition coefficient
- Molecular size and shape
