Pepsin is produced in the stomach by gastric glands. It acts in an acidic medium.

Trypsin is produced by the pancreas which acts in an alkaline medium.

Pepsin hydrolyzes peptide bonds between large hydrophobic amino acid residues, whereas trypsin  hydrolyzes peptide bonds at the C-terminal side of lysine or arginine.
