The disease is the right answer.

The virgin soil epidemic is an epidemic in which a population at risk have had no prior touch with the illness that strikes them and is, therefore, immunologic-ally nearly vulnerable.

After the arrival of Cortes in Mexico, another army came for Cortes inspection for the management. In return to this Cortez fought against the army from Cuba and there an African slave had a disease known as smallpox. And that slave came into contact with others. This disease, therefore, spread more quickly and became a reason for the depopulation of the Native American population in the 16th century.
