there is not a single difference in meaning of these two sentences
the only difference is that don't is used in AMERICAN english while do not in BRITISH english

Interrogative means when a question is asked in a sentence.
But, in a negative sentence there is negativity,i.e.do not or not.
Don't I speak english?is interrogative.
Do I not speak english?is both interrogative and negative.
