Cheek Cells Under the Microscope. Requirements, Preparation and Staining. Cheek cells are eukaryotic cells (cells that contain a nucleus and other organelles within enclosed in a membrane) that are easily shed from the mouth lining. It is therefore easy to obtain them for observation.

To observe the human cheek cells

Materials required - Microscope , slide , spoon , saffrain , or methylene blue

Procedure -

  • Wash the mouth clean
  • Take a clean plastic spoon and the scrap of inner lining of cheek
  • Take the scrapping and put it in a drop of water on the slide
  • Add a drop of methylene blue for staining
  • Cover the slide with cover slip and observe under microscope
  • Wipe the extra stain using filter paper

Observation - human cheek cell with the nucleus in the centre of cell can be observed

Inference - Human cheek cell consists of nucleus , cytoplasm and the cell membrane

Precautions -

  • Wash the spoon thoroughly before using it
  • Do not scrap it hard as it may cause injury to buccal cavity
