Explanation:

1 Denauration-unwinding the double helix by heating to 95 degrees celsius for 30 seconds

2.Annealing -priming the DNA by cooling the test tube 250 degree Celsius for 30 seconds

3.Extension-adding on complementary nucleotides and reheating to 72 degrees celsius for 60 seconds

Thermal cycler is a laboratory Apparatus most commonly used to amplify segments of DNA . thermal cycles may also be used in Laboratories to facilitate other temperature-sensitive reactions

Answer:

PCR stands for Polymerase chain reactio . It was discovered by Kary Mullis. It is used for the  amplification of DNA.

Sub- topic :-

Polymerase chain reaction is used for amplification of DNA that make billion copies of DNA in just one hour . It is widely used for detection of HIV and used for gene therepy.

Explanation:

The polymerase chain reaction has 3 steps denature, annealing and extension.

Denature :-  It is the first stage of PCR in which ds DNA is separated into two single strands by breaking hydrogen bond . It is done on 94 degree celsius .

Annealing :-  Annealing of PCR occurs at 50 to 60 degree Celsius. In which the primer base pair of a complementary region flanking DNA strands ,which is done by use of oligonucleotide sequence.

Extension:- It is done by the use of thermostable enzyme , thermus aquaticus, where the primer is added to the base pair .

Hence the DNA is amplified to 1 billion time after 30 cycle .
