1: The rising level of water in the bent tube shows that a partial vacuum was created in the flask, so the water starts moving from High pressure to low pressure area i.e. to the flask. Now the vacuum was created in the beaker due to presence of KOH which absorbs the CO2 produced by Wet and germinating seeds during respiration.

2:Boiled seeds can't be used as they don't produce CO2.
