Tween animation means the software will fill up the frames between two key frames.
Frame-by-Frame means you will have to do it.
Example :

In Frame 1 you draw a square and in Frame 20 you change that square to a triangle. Now Flash decides to fill up the in between frames from 1 to 20 such that when you play your animation, you will see a gradual smooth transformation of a square to a triangle. This is tween.
But if you decided not to use the tween feature, you will see the same square for the first 19 frame and in the 20th frame, it will suddenly change to a traingle. Then you might have to create frames at 5, 10 and 15 to give the transformation some smoothness. This is frame by frame.
