Given vertices of Square are A(-1,1) and B(5,3).

Mid-point of given diagonal = (-1+5/2,1+3/2)

                                               = (4/2,4/2)

                                               = (2,2) = (x1,y1)

The Equation of other diagonal:

y - y1 = m(x - x1)

y - 2 = -1/y2 - y1/x2-x1(x - 2)

y - 2 = -1/3 - 1/5 + 1(x - 2)

y - 2 = -1/2/6(x + 2)

y - 2 = -1 * 6/2(x + 2)

y - 2 = -6/2(x + 2)

y - 2 = -3(x + 2)

y - 2 = -3x + 6

y = - 3x + 6 + 2

y = - 3x + 8

(or) 

y + 3x - 8 = 0

(or) 

y - 8 = -3x.
