• Take a rheo leaf,tear it in a single stroke.
• observe it against the light.
• take a small piece of leaf peel with light coloured (transparent)portion.
• Put it on slide and add 1 to 2 drops of dilute salt solution on the membrane.
• Leave it for about 5 to 10 mins and observe it under the microscope.
• Due to this water from the rheo leaf cell comes out. It results in shrinkage of the cytoplasm along with cell membrane.
