For manual reticulocyte counts, whole blood is mixed with a supravital dye such as new methylene blue and blood smears are prepared. The dye causes clumping and staining of residual nucleic acid present in immature cells. The stained cells (reticulocytes) are counted as a percentage of total red blood cells

Principle for automated measurement of reticulocytes

The cells in HFR, MFR and LFR fractions6 are counted as reticulocytes. The reticulocytes in HFR and MFR fractions are counted as IRF7 (Immature Red Cell Fraction). The cells in RBC fraction are counted as mature erythrocytes
