Procedure
. Take a clean slide and put a drop of water on it in the centre.
Take inner leaf of an onion. Cut some about 1/4 inch square pieces on its inner side and snap it
backwards.
With the help of forceps, peel off the thin square of epidermal layer and transfer it onto the glass
slide having a drop of water.
Cut the membrane into a rectangular piece with the help of a sharp blade.
Drain off water and add a drop of Safranin stain and leave it for 2 minutes.
Transfer the stained peel into a watch glass containing clear water to wash off extra stain.
Put a drop of glycerine in the centre of slide and with the help of a brush and needle, place the pee
on glycerine drop on the slide. Take care that there is no fold in the peel.
Cover the peel gently with the coverslip using a needle to avoid the entry of air bubbles.
Gently, press the coverslip with the blunt end of needle to spread the glycerine evenly.​
