Not really, for two reasons:

1.    To the best of our knowledge, isolated quarks cannot be observed; they always appear as quark-antiquark pairs or quark triplets, with integer charges

2.   The fact that charge is quantized does not tell you how much the fundamental quantum of charge is. All you need to do is to assume that the fundamental quantum of charge is 1/3 e and all charges will be integral multiples!
