If my assumption is correct, the question applies to a pair of stereo speakers and 3.5mm audio jack connection. I.e. no optical output and/or 5.1 home theater etc. configuration, no dedicated (internal or external) audio device involved.

Then it mostly depends on the audio content to be mostly played (audio communications or entertainment), speakers’ quality, where would you like to place them and whether your monitor has an HDMI port.

Of course the direct connection to your audio adapter is better from the quality perspective.

On the other hand, HDMI allows you to better organize your cables using one cable to transmit the video and audio signals from your desktop PC versus 2 (HDMI+audio cables) to your monitor and then to your speakers from the 3.5mm audio jack on your monitor.

If there is no HDMI port on your monitor, then I see no real sense in connecting your PC 3.5mm audio out to the monitor’s line in and then your speakers to your monitor’s 3.5mm audio out.
