The role of nicotinamide in DNA repair and maintenance of genomic stability is tightly related to its functions as an precursor and a substrate for PARP-1. PARP-1 is a nuclear enzyme which detects DNA damage, binds to DNA single or double strand breaks, and then uses as a substrate to form nicotinamide and ADP-ribose.
