Very little was discovered about the cell for the next 150 years after Hooke's discovery in 1665.

With the development of improved microscopes and more effective staining methods in subsequent years years, it became possible to know more about cell structure and its functions.

because before 1940,scientists were observing cells under light microscopes by which only cell wall, cell membrane, vacoule, nucleus and mithochondria can be observed . The discovery of electron microscope by Knoll and Ruska in 1940 made it possible to observe the complex structure of the cell and its various cell organelles.i.e.its ultrastructure.Ribosomes,ER,Golgi apparatus, lysosomes, plastids can only be observed with an electron microscope .
