Shylock is speaking to the Duke of Venice.

The purpos-e here is a trial for the bond between Shylock and Antonio, in whic-h Shylock had lent out three thousand ducats for three months and the pe-nalty was to extract a pound of Antonio's flesh if he fails to repay. Since the bond had expired and Antonio could not pay back the amount, Shylock d-emand-ed the pe-nalty to be carried out.

Holy Sabbath is the 7th day of the holy week according to Jewish scriptures. It is a religious holiday for the Jews.

Shylock said that he is d-etermined to have the penalty since he has pledged in the name of his religion. If the Duke d-enied him what he d-es-erves rightfully, then that would jeopardize the municipal c-harter and the freedom of the country they lived in, that is Venice. This was directly intend-ed towards the core of Venice therefore it was a high scale threat.
