1. First remove the stomatal covering of the leaf carefully.

2. Then, colour it by adding saffranin for better observation.

3. Clean leaf peel if extra amount of saffranin is added.

4. Then keep it on a glass slide and add few drops of glycerine to it.

5. Finally, cover it with a cover-slip.
