AIM- to observe lactobacilli in curd buttermilk.

requirements: curds, yoga or buttermilk, glass slides ,coverslips, 1% methylene blue solution, bottling paper, microscope ,etc.

procedure: 1) take a very small drop of curd on the glass slide. with the help of a toothpicks smear it continuously until a film of the curd is formed on the slides.

2) led the smear dry.

3) place a small drop of methylene blue solution on a microscope slide. this is training the slide.

4) place a cover slip on top. remove excess solution around the cover slip with a blotting paper.

5) place this slide on the stage of the microscope starting with the lower magnification. after observing the bacteria under low power as tiny dots, shift the eyepiece to the high power magnification.

Conclusion: lactobacilli are bacteria belonging to the kingdom Monera. lactobacilli produce lactic acid from the fermentation of carbohydrates. they therefore carry out curdling process of the milk.

observations : the lactobacilli are rod shaped bacterium. they do not show nuclei aur cell organelles. they are prokaryotic cell.
