CSC Theory originated with a rare gonadal malignancy termed an embryonal carcinoma (EC). As far back as the late 1800s, pathologists had described ECs as a disorganized caricature of the tissues (endoderm, mesoderm, and ectoderm) of an early stage embryo. This led to the postulation that ECs may develop from a pluripotent cell in a parallel fashion to the embryo. By the 1960s, single EC cells (ECCs) had been isolated and shown to be capable of reproducing their original malignancy in xenograft models [11]. By the 1980s, pluripotent ECCs had been developed into what was in effect the first CSC cell culture model [12–16], although the “CSC” term was not used at the time. Cells from a well-differentiated EC termed a “teratocarcinoma” could be maintained in the SR state in culture and readily differentiated by the addition of all-trans retinoic acid (ATRA) [16]. In parallel, bone marrow research had attracted substantial interest following the devastating effects of the 1945 nuclear attacks in Japan. Substantial progress was made in understanding bone marrow stem cells (HSCs and MSCs), which ultimately led to the identification of Leukemia SCs (LSCs) in 1997, when the term “CSC” was coined [17,18]. At this point, however, it was not clear whether CSCs were involved in other malignancies.
