the illustration shows a girl pushing on heavy box. She pushes with a force of 40 N. How can you determine if she is doing work on the box?

Well, if the box moves, she is doing some work otherwise, she is not doing any work.

According to physics work is only said to be done if a force is applied and the object shows some displacement.
If the box pushed by the girl shows any displacement, then only it can be said that work is done.
W=F.S (dot product of vectors)
will be the formula to calculate work done.
