Bindusara wanted his son Susima to succeed him however, Ashoka was the unanimous choice among his father's ministers, who found their Emperor's choice to be arrogant and disrespectful towards them.
Royal Minister Radhagupta seems to play a vital role in pages of history and reason behind Ashoka's rise to the throne. He was later appointed as prime minister, once Ashoka seated on throne.
According to Mahavansa and Dipavansa, Ashoka killed 99 of his brothers, sparing only one, named Vitashoka or Tissa or Tishya.
Please note that there is no clear proof about this incident (as mentioned in wikipedia to it is more or less saturated with mythological elements).
