Basically, gene cloning involves the following steps:

  • The gene of interest is that organism's DNA is extracted and cut into the size of genes with the help of restriction enzymes.
  • The bacterial plasmids (DNA) are also cut with the same enzyme.
  • Both cut DNA and plasmid are kept together in a test tube. Their combination gives rise to recombinant DNA.
  • The recombinant DNA is transferred into bacteria using heat shock.
  • The bacteria multiply into colonies called gene libraries.
  • The gene library is then screened to identify the gene of interest by observing three things on each i.e a sequence of DNA similar to the gene of interest, a protein encoded by a gene of interest, a DNA marker with a location close to a gene of interest.
