- Preparation of sterile environmental conditions to prevent the growth of microbes in the cell culture.
- Preparing culture media according to the growth requirements of the cells. Agar is generally added as a solidifying agent
- Inoculating the mother plant in the medium.
- Then this explant is incubated under adequate conditions of temperature.
- Callus (i.e. tissue growth) is observed on the medium.
- Organogenesis takes place involving dedifferentiation and redifferentiation.
- The complete organism of interest has developed from the explant.
