They have known each other since their school days.

In English linguistics, we use since when depicting an event which began in the past and continues to occur in the present.
On the other hand, we use 'for' for a specified period of time.
As the question statement depicts they they started to know each other in the past and still know each other in the present, hence since will be used to fill in the blank.
