Principle:

In this experiment, moistened gram seeds are used. So that they release carbon-di-oxide and anaerobic respiration occurs. Anaerobic respiration is nothing but it lacks oxygen. KOH solution consumes the released carbon-di-oxide.

Method

  • Germinate seeds by wrapping them in moist blotting paper or cotton wool for around 3 to 4 days.
  • Place the germinated seeds in a conical flask and some water is sprayed into the flask. Use the thread to suspend the conical flask that has the test tube with 20% KOH solution. The rubber cork is used to seal the flask opening.
  • In this case,  insert one edge of the U-shaped glass delivery tube through the hole in the rubber cork and one edge into a beaker saturated with water. It is important to note that all the attachments are sealed.
  • It is necessary to mark the initial level of water in the U-shaped delivery tube.
  • The experiment is left uninterrupted for 1 to 2 hours to observe the water fluctuation in the test tube.

Observation and Conclusion :

  • During the respiration process in the conical flask, carbon dioxide is released from the germinating gram seeds, which indicates the rising level of the water.
  • The KOH solution suspended in the test tube in the conical flask absorbs or consumes the carbon dioxide released during the process, resulting in a vacuum or void in the flask and upward water movement in the test tube. As a result, the water level in the test tube fluctuates.

Precautions

  • Moisturizing seeds before germinating them is necessary
  • All connections in the experimental arrangement must be kept in an airtight environment.
  • The KOH solution used must be freshly prepared.
  • One end of the delivery tube must be kept in the conical flask. The beaker's other edge is submerged in water.
