2. TYPES OF SUPPOSITORY Suppositories are prepared by three methods:- • Molding • Compression • Hand rolling and shaping

3. MOLDING Melting the base Incorporating any required medicament Pouring the melt into molds Allowing the melt to cool and congeal into suppositories Removing the formed suppository from the mold The steps in molding include :-

4. EXAMPLES •Cocoa butter •Glycerinated gelatin •Polyethylene glycol

5. ADVANTAGES • It is a simple process • More elegant than hand rolling • No solid sedimentation in the base

6. Compression:- Base and other formulative ingredients are mixed Suppository mass is placed in a cylinder The cylinder is closed,pressure is applied on one end When mold is filled ,movable end plate is removed The formed suppositories are ejected The steps are:-

7. ADVANTAGES • It is a simple method. • It gives suppositories that are more elegant than hand molded suppositories. • In this method sedimentation of solids in the base is prevented. • Suitable for heat labile medicaments.

8. DISADVANTAGES • Air entrapment may take place. • This air may cause weight variation. • The drug and/or the base may be oxidized by this air.

10. HAND ROLLING AND SHAPING The drug is made into fine powdef Incorporated into supoository base By trituration and kneading Mass is rolled into rod shaped by fingures Rods are cut into pieces Desired shape is given by hands • It is the oldest and simplest method. The steps involve :-

11. ADVANTAGES • It has the advantage of avoiding the necessity of heating the cocoa butter • No heating • No Mold Preparation

12. DISADVANTAGES • Manupilation requires considerable skill • Appearance is not elegant
